چکیده

In Alzheimer`s disease (AD), regional heterogeneity of ?-amyloid burden and microglial activation individual patients is a well-known phenomenon. Recently, we described high incidence inter-individual in terms asymmetry plaque mouse models AD as assessed by positron-emission-tomography (PET). We now investigate the associations between amyloid burden, activation, impaired spatial learning performance transgenic mice vivo. 30 AppNL-G-F (15 female, 15 male) acquired cross-sectional 18 kDa translocator protein (TSPO-PET, 18F-GE-180) ?-amyloid-PET (18F-florbetaben) scans at ten months age. Control data were obtained from age- sex-matched C57BI/6 wild-type mice. (i.e. Morris water maze) within two weeks PET scanning correlated principal component scores with voxel-wise TSPO tracer uptake maps mice, controlled for age sex. order to assess effects hemispheric asymmetry, also analyzed correlations bilateral regions interest frontal cortex, entorhinal/piriform amygdala, hippocampus, using regression model. tested correlation biomarkers performance. Voxel-wise analyses revealed that higher TSPO-PET signal entorhinal piriform cortices, hippocampus hypothalamus Region-based analysis showed significant expression right cortex amygdala performance, whereas there no such left hemisphere. Right lateralized predicted better maze (? = -0.470, p 0.013), irrespective global level. results amyloid-PET learning. Elevated amygdala-entorhinal-hippocampal complex associated Our findings support protective role microglia on cognitive function when they highly express specific brain involved memory.

Age-associated dysregulation of microglial activation is coupled with enhanced BBB permeability and pathology in APP/PS1 mice

Running title: altered microglial activation states in aged APP/PS1 mice 2 Abstract Aging adversely affects inflammatory processes in the brain, which has important implications in the context of disease progression. It has been proposed that microglia become dysfunctional with age and may lose their neuroprotective properties leading to chronic neurodegeneration. Here, we sought to characteriz...

Amyloid-beta deposition is associated with decreased hippocampal glucose metabolism and spatial memory impairment in APP/PS1 mice.

In Alzheimer disease (AD) patients, early memory dysfunction is associated with glucose hypometabolism and neuronal loss in the hippocampus. Double transgenic (Tg) mice co-expressing the M146L presenilin 1 (PS1) and K670N/M671L, the double "Swedish" amyloid precursor protein (APP) mutations, are a model of AD amyloid-beta deposition (Abeta) that exhibits earlier and more profound impairments of...

Differential effects of inactivation of the right and left basolateral amygdala on spatial memory in place avoidance task in rats

There is strong evidence that two cerebral hemispheres are differentially involved in emotional memory and that amygdala is a key subcortical structure for emotional experience. The present research investigated the possible involvement of lateralization of basolateral amygdala (BLA) and central amygdala (CEA) in place avoidance memory. For this purpose, male Long-Evans rats (280-320 g) were im...

Hippocampal dysregulation of neurofibromin-dependent pathways is associated with impaired spatial learning in engrailed 2 knock-out mice.

Genome-wide association studies indicated the homeobox-containing transcription factor Engrailed-2 (En2) as a candidate gene for autism spectrum disorders (ASD). Accordingly, En2 knock-out (En2(-/-)) mice show anatomical and behavioral "ASD-like" features, including decreased sociability and learning deficits. The molecular pathways underlying these deficits in En2(-/-) mice are not known. Defi...

Enhanced visual cortical activation for emotional stimuli is preserved in patients with unilateral amygdala resection.

Emotionally arousing pictures induce increased activation of visual pathways relative to emotionally neutral images. A predominant model for the preferential processing and attention to emotional stimuli posits that the amygdala modulates sensory pathways through its projections to visual cortices. However, recent behavioral studies have found intact perceptual facilitation of emotional stimuli...
